In two GLP tests according to OECD 301F and OECD 301C using non-adapted municipal sludge and mixed inoculum no bodegradation was measured after 28 days of exposure (BASF 1999, BASF 2013). In order to clarify whether the substances is persistent or not a GLP test according to ISO 9439 was set up also using non-adapted municipal sludge as inoculum but with a prolonged contact time of 60 days (BASF 2011). It was found that the substance has a lag phase of ca. 21 days. After this lag phase the degradatiom starts slowly. The degradation curve is never sigmoid. After 60 days the degree of biodegradation is approx. 30% related to the formation of carbon dioxide.

The model CATALOGIC 301C v11.15 (OASIS Catalogic v5.13.1.156) calculated biodegradation of 3 -API to be 56% after 28 days, based on CO2 -Evolution. Tthe substance was within the model’s applicability domain.

Metabolites:

The model Catalogic 301 C v11.15 (OASIS Catalogic v5.13.1.156) calculated 44 metabolites identifying 7 metabolites as relevant degradation with an estimated quantity of≥ 0.1% (substance the applicability domain; for details see ‘Attached background material’ of the respective Endpoint Study Record). One out of seven metabolites identified as relevant degradation products were calculated to be not readily biodegradable. Individual biodegradation of this metabolite was estimated to be 53% after 28 days (based on BOD).

Conclusion:

Summing up the available experimental and calculated data, 3 -API (CAS 5036 -48 -6) is concluded via weight-of-evidence approach as inherently biodegradable showing biodegradation after a prolonged exposure period and a prolonged lag phase.
