Since no reliable chronic fish test is available for C12-16AS Na (CAS 73296-89-6), a read-across to another structurally related category members was conducted in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5. Read-across data from C12AS Na (CAS 151-21-3) and C14-15AS Na (CAS 68081-98-1) were used. Toxicity of alkyl sulfates is dependent on the carbon chain length.
A weight of evidence approach is performed using experimental data of three chronic fish studies available for alkyl sulfate sodium salts. In two of the studies fathead minnows (Pimephales promelas) were exposed to a C14-15AS Na (CAS 68081-98-1). In the remaining study the pure homologue C12AS Na (CAS 151-21-3) was tested on the same fish species. As the C14-chain length is regarded to be the most toxic in chronic tests on aquatic organisms (OECD SIDS, 2007), the C14-15AS Na (CAS 68081-98-1) is expected to be even more toxic than the C12-16AS Na (CAS 73296-89-6). Thus the read-across is a worst-case assumption and therefore studies on the C12-homologues were additionally evaluated.
In the non-GLP study of Sousa (P&G, 1987), which was conducted equivalent to the OECD guideline 210, fathead minnow was exposed to C14-15AS Na over 34days in a flow-through system. The early-life stage test resulted in a NOEC-value of 0.35 mg/L for growth (length/weight). This result corresponds well with the NOEC-value determined in a GLP-study of Versteeg and Rawlings (P&G, 1997). In this test fathead minnows (Pimephales promelas) were exposed with the same mixture in an outdoor flow-through system over 35 days. The exposure method is described as equivalent to OECD 53, a guidance on simulated freshwater lentic field tests. The resulting NOEC-value is reported to be 0.371 mg/L for growth (for length and weight).
By testing the pure homologue substance C12AS Na (CAS 151-21-3) a higher NOEC-value is reported by Belanger (1995). In the publication of Belanger the NOEC for weight of larvae of Pimephales promelas was reported to be >= 1.357 mg/L after exposure for 42 days in an outdoor flow-through system.
As the C12-16AS Na contains relatively high amounts of C12 (60 -71%), toxicity for that mixture is expected to be even lower than reported for the mixture C14-15AS Na. The content of the toxic C14 vary between 20 and 30%. The remaining alkyl sufate homologue of the composition C16 (4 -15%), is expected to be lower toxic than the C14 AS homologue (OECD, SIDS, 2007).
