- - Test water was sampled and pH, hardness, conductivity and alkalinity were measured by the study laboratory. Water quality analysis including DOC, COD, suspended solids, N, pesticides, PCBs and metals was conducted in a separate non-GLP laboratory. Metal analysis of four test solutions (3.2 mg/l, 56 mg/l and two controls) were conducted in another non-GLP laboratory.

- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: The test solutions were prepared by the addition of a known quantity of mixed ash directly to 1 litre volumes of culture medium. These solutions were stirred overnight using magnetic followers and then allowed to stand for 15 minutes. After this time the solutions were filtered through 0.45 µm cellulose nitrate filters, which had been previously washed with 100 mL Reverse Osmosis (RO) water. The pH of each solution was measured and adjusted, if necessary, with 1 M hydrochloric acid to 8.0 ± 0.25. The resulting solutions were clear and colourless.

- Executive summary:
- Short-term toxicity of Ash was determined in a GLP compliant study according to OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test). In the test, young, less than 24 h old D. magna were exposed to the test substance for 48 hours in a static test. Due to low water solubility of Ash, test solutions were filtrated to form Water Accommondable Fractions (WAF) of Ash. Immobilisation was observed 24 and 48 hours after initiation of the test.The EC50 value was determined to be 57.1 mg/L.

Physical and chemical data
Dissolved oxygen concentrations ranged from 6.2 to 9.2 mg/L (68 to 101% of the air saturation value), pH values (after time 0 adjustments) ranged from 7.1 to 8.2, and the salinity ranged from 0.2 to 0.3‰. Individual data are shown in Table 5.

Temperature values taken at 0, 24 and 48 hours were 21.6, 21.4 and 21.1°C.
The hardness of the D. magna media was 181 mg/L CaCO3, and the conductivity was 679 µS/cm. Water quality parameters of a recent batch of D. magna media are shown in Table 6.
