No study investigating the short-term toxicity of Monopentaerythritol tetraesters and dipentaerythritol hexaesters of valeric, heptanoic and nonanoic acids to freshwater fish is available. Therefore, in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5 a read-across to the structurally related source substances Decanoic acid, mixed esters with heptanoic acid, octanoic acid, pentaerythritol and valeric acid (CAS 71010-76-9), Tetra-esterification products of C5, C7, C8, C10 fatty acids with pentraerythrito (CAS 68424-31-7), Carboxylic acids, C5-9, tetraesters with pentaerythritol (CAS 67762-53-2), Fatty acids, C8-10 mixed esters with dipenaterythritol, isooctanoic acid, pentaerythritol and tripentaerythritol (CAS 189200-42-8) and 2,2-bis(hydroxymethyl)-1,3-propanediyl dioleate (CAS 25151-96-6) is applied.
Based on the high degree of similarity between the structural and physico-chemical properties of the target and source substances, the source substances are considered suitable representative for the evaluation of the short-term toxicity of the target substance to fish. The read-across approach is justified in detail within the analogue justification in IUCLID section 13.
The first study investigated the short-term toxicity of the source substance Decanoic acid, mixed esters with heptanoic acid, octanoic acid, pentaerythritol and valeric acid (CAS 71010-76-9) to freshwater fish according to OECD 203 under static conditions using Brachydanio rerio as test organism. Nominal test concentrations of 100 and 150 mg/L were applied. Test substance monitoring using TOC analysis measured test concentrations of 0.696 and 0.704 mg/L, respectively. The 96 h-LC50 was determined to be > 0.704 mg/L (arithmetic mean).
The study with the source substance Tetra-esterification products of C5, C7, C8, C10 fatty acids with pentraerythrito (CAS 68424-31-7) investigated the short-term toxicity to Salmo gairdneri according to OECD 203 under semi-static conditions. Fish were exposed to a limit concentration of 1000 mg/L (nominal) for 96 hours. No mortality was observed within the tested concentration (1000 mg/L nominal) during exposure.
The short-term toxicity of the source substance Carboxylic acids, C5-9, tetraesters with pentaerythritol (CAS 67762-53-2) to freshwater fish was conducted according to OECD 203 under static conditions using Cyprinus carpio as test organism. A nominal test concentration of 100 mg/L (prepared as WAF) was applied. Test substance monitoring measured a concentration of 4.8 mg/L. No mortality was observed in the treatment throughout the test period of 96 h. Hence, the 96 h-LL50 was determined to be > 100 mg/L (nominal) and > 4.8 mg/L (measured).
The short-term toxicity of Fatty acids, C8-10 mixed esters with dipenaterythritol, isooctanoic acid, pentaerythritol and tripentaerythritol (CAS 189200-42-8) to fathead minnow was tested according to EPA OTS 797.1400 under semi-static conditions. Fish were exposed for 96 hours to nominal concentrations of 0.312, 0.625, 1.25, 2.5 and 5.0 mg/L corresponding to initial measured concentrations of 0.241, 0.847, 1.30, 2.15 and 4.11 mg/L, respectively. No mortality was observed in the treatments throughout the test period of 96 h. Hence, the 96 h-LC50 was determined to be > 5.0 mg/L (nominal) and > 4.11 mg/L (measured).
The toxicity of 2,2-bis(hydroxymethyl)-1,3-propanediyl dioleate (CAS 25151-96-6) to freshwater fish was investigated according to ISO 7346-1 under semi-static conditions using Brachydanio rerio as test organism. Nominal concentrations of 3000 and 10000 mg/L were applied. No mortalities occurred after the test duration of 96 hours resulting in an LC50 of > 10000 mg/L (nominal).
Based on the available results from structurally related source substances (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5) which are characterized by a similar ecotoxicological profile and comparable structure, it can be concluded that Monopentaerythritol tetraesters and dipentaerythritol hexaesters of valeric, heptanoic and nonanoic acids will not exhibit short-term effects to fish up to the limit of water solubility.
