Sampling and analysis
- Analytical monitoring:
- yes
- Details on sampling:
- The incubation period was 72 hours, biomass was measured in samples by counting the number of cells using a miscroscope at 0, 24, 48, and 72 hours. Before the test start pH was measured in all solutions, and again in all replicates after 72 hours.
Test solutions
- Vehicle:
- no
- Details on test solutions:
- Fresh stock solution was prepared on the day of the test start. 1000 mg of the test item was added to 1000 ml reversed osmosis water and stirred for 10 minutes. To remove the non-water soluble fraction of the test item from the test solution, the solution was filtered through a 0.45 um cellulose acetate filter (pre-boiled in order to remove any traces of external carbon). The acquired water phase (loading rate 1000 mg/l) was then used as a stock solution, from which the test concentrations were diluted using growth media. Growth media alone was used as the control solution. Nominal loading rates tested were 28, 56, 113, 225, 450 and 900 mg/ l.
Test organisms
- Test organisms (species):
- Raphidocelis subcapitata (previous names: Pseudokirchneriella subcapitata, Selenastrum capricornutum)
- Details on test organisms:
- The test organism is a laboratory culture of the green alga Pseudokirchneriella subcapitata. The inoculum is taken from Toxicon's culture of P. subcapitata (NIVA CHL 1), which originates from NIVA, Oslo, Norway. A qualification test on the alga culture was performed on the 27th of June 2013 in order to assure the quality of the test organism using potassium dichromate. ErC50 was 0,93 mg/ l, which confirmed that the culture was suitable as a test organism according to OECD 201.

Applicant's summary and conclusion
- Validity criteria fulfilled:
- yes
- Conclusions:
- The toxicity of Capa 3050 to the freshwater algae Pseudokirchneriella subcapitata was determined in a 72 hour static test. Algae were exposed to nominal loading rates of Capa 3050 between 28 and 900 mg/l for 72 hours. The 72 hour static ErC50 was 490 mg/l.
- Executive summary:
The purpose of the test was to determine the effects of Capa 3050 on the growth of Pseudokirchneriella subcapitata. Exponentially growing cultures of Pseudokirchneriella subcapitata were exposed to Capa 3050 over a period of 72 hours with sampling at 0, 24, 48, and 72 hours. OECD Guidelines 201 were followed for this test. The response was measured as the reduction of growth in a series of loading rates (28, 56, 113, 225, 450 and 900 mg/ l). Stability of Capa 3050 was confirmed in the test media. The 72 hour ErC50 was 490 mg/l based on nominal loading rates.
