The study was performed to assess the effect of the test item on the growth of the green alga Desmodesmus subspicatus following OECD Guideline 201. Following a preliminary range-finding test, the alga was exposed to an aqueous solution of the test item at a concentration of 0.32 mg/L (six replicate flasks) for 72 hours, under constant illumination and shaking at a temperature of 24±1ºC. Exposure of Desmodesmus subspicatus to the test item gave EC50 values of greater than 0.32 mg/L and correspondingly the NOEC was 0.32 mg/l. The test concentration of 0.32 mg/l was the highest attainable test concentration that could be prepared due to the limited solubility of the test item in water and auxiliary solvent, and having due regard to the amount of auxiliary solvent permitted in the test under the OECD Guidelines. Based on the geometric mean measured test concentrations, exposure of the alga to the test item gave EC50 values of greater than 0.065 mg/l and correspondingly the NOEC was considered to be 0.065 mg/l, the maximum attainable concentration under the conditions of the test. However these were a result of unavoidable loss of test substance due to abiotic (and possibly biotic) degradation of the test substance under the conditions of the assay and as no effects were observed cannot be used to derive an EC50 or a NOEC.

A decline in measured test concentrations was observed at 72h to less than the limit of quantitation (LOQ) of the analytical method employed which was determined to be 0.023 mg/L. This decline in measured test concentrations was in line with the preliminary stability analyses conducted which indicated that the test item was unstable over the test period, particularly in the light
Current regulatory advice is that in cases where a decline in measured concentrations is observed, geometric mean measured concentrations should be used for calculating EC50 values. However, no EC50 was observed in this case.
