The three acute tests on the three trophic levels are available on Reactive Orange 16 and on similar substance 1.

Reactive orange 16 was administered to zebrafish in fresh water at concentration of 500 mg/L for 96 hours according to the method described in OECD 203. No mortality and no symptoms were observed . The test substance was stable throughout the experimental period. (Markert M. Weigand W. 1985).

The acute toxicity on Daphnia  was tested on similar substance 1 by OECD 202 for 24 hours and the result is EC 50 > 304.235 mg/L.

The test on alga is available for Reactive Orange 16 and the real toxic effect has demonstrated that the observed growth inhibition effect of the test substance on Scenedesmus subspîcatus was caused in a high degree (> 50 %) due to the indirect effect, the light absorption in the coloured test solutions. Thus, the 72-hours EµC 50 and EbC 50 for Reactive Orange 16 could be determined to be clearly higher than 320 mg test substance/L when based on the real toxic effect.

There is no toxicity on microorganims, based on a study  available on Reactive Orange 16, with a contact time of 30 minutes. At the highest tested concentration :100 mg / L , no significant inhibition was observed. Same results were confirmed by a test on similar substance 2, toxicity to bacteria ("bacterial toxicity") in the oxygen-inhibition test ("test Robra") plants with activated sludge from municipal wastewater biological treatment Frankfurt / M - Sindlingen as inoculum. The provision of the inhibition rate of oxygen consumption after 3 hours of exposure to the test item on bacteria test gave the following results:

Median inhibitory concentration, EC50: >> 1000 mg/L , NOEC, EC20 : >1000 mg/L (655 mg/L based on active ingredient) , EC 80: >> 1000 mg/L.

On soil and sediment no tests are conducted because the exposure is unlikely.
