PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
A nominal amount of test item (100 mg) was dissolved in test water and the volume adjusted to 1 liter to give the 100 mg/L test concentration. The prepared concentration was inverted several times to ensure adequate mixing and homogeneity.
Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: daphnia magna
- Strain/clone: first instar Daphnia magna derived from in-house laboratory cultures
- Adult daphnids were maintained in 150 mL glass vessels containing 100 mL Elendt M4 medium in a temperature-controlled room maintaining the water temperature at 18 to 22 °C. The lighting cycle was controlled to give a 16 hours light and 8 hours darkness cycle with 20-minute dawn and dusk transition periods. Each culture was fed daily with a mixture of algal suspension (Raphidocelis subcapitata) and GEMMA Micro 300 fish food suspension. Culture conditions ensured that reproduction was by parthenogenesis. Gravid adults were isolated the day before initiation of the test, such that the young daphnids produced overnight were less than 24 hours old. These young daphnids were removed from the cultures and used for testing. The diet and diluent water are considered not to contain any contaminant that would affect the integrity or outcome of the study.

METHOD FOR PREPARATION AND COLLECTION OF EARLY INSTARS OR OTHER LIFE STAGES, INCLUDING CULTURING CONDITIONS:
Gravid adults were isolated the day before initiation of the test, such that the young daphnids produced overnight were less than 24 hours old. These young daphnids were removed from the cultures and used for testing.
Test type:
static
Water media type:
freshwater
Limit test:
yes
Total exposure duration:
48 h
Hardness:
2.5 mmol/L (= 250 mg/L as CaCO3)
Test temperature:
21°C
pH:
7.9+/-0.3
Nominal and measured concentrations:
Analysis of the test preparations at 0 and 48 hours showed measured test concentrations to range from 103% to 104% of nominal respectively and so the results are based on the nominal test concentration only.
Details on test conditions:
TEST SYSTEM
- Test vessel: 120 mL glass vessels
- Material, size, headspace, fill volume: Each 120 mL test and control vessel was completely filled with test media and sealed to reduce evaporation.
- Volume of solution: 120 ml
- Aeration: no
- No. of organisms per vessel: 5
- No. of vessels per concentration (replicates): four replicate test vessels
- No. of vessels per control (replicates): four replicate control vessels

TEST MEDIUM / WATER PARAMETERS
Reconstituted Water- Elendt M4 Medium

OTHER TEST CONDITIONS
- Adjustment of pH: If necessary the pH may be adjusted with NaOH or HCl.
- Photoperiod: photoperiod of 16 hours light and 8 hours darkness for a period of 48 hours with 20-minute dawn and dusk transition periods
- Light intensity: between 200 and 1200 Lux

EFFECT PARAMETERS MEASURED:
Any immobilization or adverse reactions to exposure were recorded at 24 and 48 hours after the start of exposure. The criterion of effect used was that Daphnia were considered to be immobilized if they were unable to swim within 15 seconds after gentle agitation.

VEHICLE CONTROL PERFORMED: no, no vehicle used

RANGE-FINDING STUDY
- Test concentrations: 0.10, 1.0, 10 and 100 mg/L.
- Results used to determine the conditions for the definitive study: No sub-lethal effects of exposure were observed throughout the test.

Duration:
48 h
Dose descriptor:
NOEC
Effect conc.:
100 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mortality
Remarks:
There was no immobilization in 20 daphnids exposed to a test concentration of 100 mg/L for a period of 48 hours. No sub-lethal effects of exposure were observed throughout the test
Details on results:
- Verification of Test Concentrations
Analysis of the test preparations at 0 and 48 hours showed measured test concentrations to range from 103% to 104% of nominal respectively and so the results are based on the nominal test concentration only.
- Immobilization Data
There was no immobilization in 20 daphnids exposed to a test concentration of 100 mg/L for a period of 48 hours.
The No Observed Effect Concentration after 24 and 48-hours exposure was 100 mg/L.
It was considered unnecessary and unrealistic to test at concentrations in excess of 100 mg/L.
- Sub-lethal effects
No sub-lethal effects of exposure were observed throughout the test
Validity criteria fulfilled:
yes
Conclusions:
The acute toxicity of the test item to the freshwater invertebrate Daphnia magna has been investigated and gave a 48-Hour EC50 value of greater than 100 mg/L. The NOEC was 100 mg/L.
Executive summary:

Introduction
A study was performed to assess the acute toxicity of the test item to Daphnia magna. The method followed was designed to be compatible with the OECD Guidelines for Testing of Chemicals (April 2004) No 202, "Daphnia sp., Acute Immobilisation Test" referenced as Method C.2 of Commission Regulation (EC) No. 440/2008.
Methods
Following a preliminary range-finding test, 20 daphnids (4 replicates of 5 animals) were exposed to an aqueous solution of the test item at a nominal concentration of 100 mg/L for 48 hours at a temperature of approximately 20 to 21 °C under static test conditions. The number of immobilized Daphnia was recorded after 24 and 48 hours.
Results
Analysis of the test preparations at 0 and 48 hours showed measured test concentrations to range from 103% to 104% of nominal respectively and so the results are based on the nominal test concentration only.
Exposure of Daphnia magna to the test item gave EC50 values based on the nominal test concentration of greater than 100 mg/L. The No Observed Effect Concentration (NOEC) was 100 mg/L.
It was considered unnecessary and unrealistic to test at concentrations in excess of 100 mg/L.

Description of key information

The acute toxicity of the test item to the freshwater invertebrate Daphnia magna has been investigated and gave a 48-Hour EC50 value of greater than 100 mg/L. The NOEC was 100 mg/L.
