- Executive summary:
The toxicity of test substance (ca. 40% a.i.) to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined according to OECD Guideline 201, in compliance with GLP. The aim of the study was to assess the effects on growth rate and yield over a period of 72 h. The study was conducted under static conditions with an initial cell density of 1 x 10E+4 cells/mL. Six concentrations were tested in a geometric series with a dilution factor of 2.2: 0.0150, 0.0340, 0.0750, 0.165, 0.364 and 0.800 mg/L of the test substance. Three replicates were used to determine alga growth inhibition whereas six replicates were used for the control without addition of test substance. Analytical dose verification was conducted using HPLC-MS. The test substance was found to inhibit growth of the freshwater green alga after 72 h with the following effect values: The EC50 values with 95% confidence intervals for inhibition of specific growth rate (ErC50) after 72 h were 0.156 (0.0981 – 0.249) mg/L. In the present report, the 96 h NOErC of 0.011 mg a.i./L was selected as key value for the derivation of aquatic PNEC values, in line with the methodology followed in the DDAC biocides assessment report for Product Type 8.

The DDAC biocides assessment report for Product Type 8 conducted under Directive 98/8/EC (evaluating Competent Authority: Italy, June 2015, attached in Section 13 of the IUCLID dataset), reported an additional study on DDAC, apart from the above EQC studies. This study resulted in a 96 h ErC50 = 0.021 mg a.i./L and a 96 h NOErC = 0.011mg a.i./L. The RMS considered the value from this study as the endpoint for the product authorisation.
